What boss gave you the most headache on your first play-through?
For me, it was the godskin noble. Fuck this guy. I was using a great-sword build. I played 20 hours of this game before I hit a wall on the godskin noble. I had to rune farm and level up an extra 15 levels before I stood a chance.
